ABSTRACT

Objective: Using the COI Barcode to establish the standard method to identify the traditional Chinese medicine of deer products. Methods: In this study, DNA extraction, PCR amplification, and sequencing of experimen-tal samples (deer antler, penis and testis, tendon, tail, bone, foetus) were successful. COI sequence database were constructed and commercial crude drugs of deer were investigated and analyzed. Results: By using universal COI primer,PCR amplification is preferably. All the species could be identified based on COI sequences database of tra-ditional Chinese medicine of deer which contained 101 samples of 8 species. We have collected 40 commercial crude drugs in which 18 samples were identified as species described in Chinese pharmacopoeia. Conclusion: DNA barcode technology based on COI sequence could be a standard approach to identify traditional Chinese medicine of deer products, and provide the basis for the identification of commercial deer medicine.

ABSTRACT

Objective: This study aimed to distinguish Serpentis Periostracum from its adulterants, which will provide the basis for its safe application. Methods: Here, COI sequences of 68 samples from 13 species were PCR amplified and sequenced. Furthermore, the DNA Barcoding Gap and phylogenetic cluster analysis were carried out. Results:The results exhibited that the COI sequences of all the three origin animals of Serpentis Periostracum have DNA Barcoding Gap. For phylogenetic cluster analysis, all the three origin animals showed monophyletic and every species can be discriminated clearly. Conclusion: COI is an effective DNA barcode for the identification of Serpen-tis Periostracum.

ABSTRACT

DNA barcoding technology is widely opplied for species identification and discovery by using short and standard fragments of DNA sequences. In this study, the cytochrome c oxidase subunit 1 (COI) sequence as a DNA barcode was used to identify the Gekkogecko Linnaeus medicinal materials and adulterants in order to provide a new identification method of G. gecko Linnaeus. Genomic DNA was extracted from the experimental samples. The COI regions of nrDNA were amplified using polymerase chain reaction and sequenced bidirectionally. The alignment and NJ tree construction were performed in MEGA6.0. COI sequences can be obtained from all samples. The average intra-specific K2P distance of G. gecko Linnaeus was 0.005 and the maximum intra-specific distance was 0.013. All the G. gecko Linnaeus medicinal samples clustered into a clade in the NJ tree and can be distinguished from the adulterants. In a conclusion, COI can be used to correctly identify G. gecko Linnaeus, and it will be a potential DNA barcode for identification of other animal medicinal materials.

ABSTRACT

Objective To explore the relationship between the rs1611115 and rs739398 polymorphism of dopamine-β-hydroxy-lase genes and violent behavior in adolescents .Methods Adolescent male offenders were divided into violent group (107 samples) and nonviolent group (107 samples) .Besides ,107 male adolescents in vocational school were served as normal controls .The rs1611115 and rs739398 polymorphisms of DβH gene were measured by used SNaPshot SNP technology in 321 samples .Chi-square test was used to compare the distribution difference between three groups of each gene type and the allele frequency distribution . Results There was statistically significant difference among violent group ,nonviolent group and normal control group in the geno-type and allele frequencies of DβH gene rs1611115 polymorphism(P0 .05) .There was no statistically significant difference among violent group ,nonvio-lent group and normal control group in the genotype and allele frequencies of rs739398 polymorphism(P>0 .05) .Conclusion No association was found between the rs739398 polymorphism of DβH gene and adolescents with violent behavior .But the rs1611115 polymorphism of DβH gene was related to adolescent criminals with violent and nonviolent behavior .

ABSTRACT

Objective To explore the relationship between the rs3785143 polymorphism of norepinephrine transporter gene, and investigate the influence of interaction effect between NET gene polymorphism and childhood abuse in adolescents with violent behavior. Methods Adolescent male offenders were divided into violent group (124 samples) and nonviolent group (120 samples). The rs3785143 polymorphisms of NET gene were measured by used SNaPshot SNP technology in 244 samples.Chi-square test was used to compare the distribution difference between three groups of each gene type and the allele frequency distribution. Results ①There was no statistically significant difference among violent group and nonviolent group in the genotype and allele frequencies of rs3785143 polymorphism (x2=0.753,0.022; all P>0.05).②64.5% violent adolescents have experience of childhood abuse, and 35.5% had no history of abuse ;48.3% nonviolent adolescents have experience of childhood abuse, and 51.7% had no history of abuse(x2=6.50,P<0.05).③There was not significant interaction between NET gene polymorphism and childhood abuse((OR=0.386,P>0.05) 95% CI (0.457-4.739)).Conclusions The NET gene polymorphism is not associated with violent behavior in male adolescents, and there was not interaction between NET gene polymorphism and childhood abuse.

ABSTRACT

Objective To analyze the investigation and treatment of signet ring cell carcinoma of pancreas.Method The clinical data of patients with histopathologically confirmed signet ring cell carcinoma of pancreas were analyzed retrospectively. Results There were 4 patients with signet ring cell carcinoma. There was no patient who presented with a typical carcinoid syndrome. Most patients presented with upper abdominal pain, backache or jaundice caused by bile duct obstruction. In one patient CA19-9 and CEA were raised. All patients received palliative biliary bypass and needle biopsy of the tumour. The median survival was 2.8 months. Conclusions Pancreatic signet ring cell carcinoma is a rare disease with poor prognosis.Surgery is the only effective treatment but the resectability rate is low.Whether this tumour responds to chemotherapy requires further studies.
